For most modern vehicles, yes, the engine should be running when you check the automatic transmission fluid. The engine needs to be at operating temperature and idling to circulate the fluid, providing an accurate reading on the dipstick. Checking it with the engine off will almost always show an incorrectly high level, which can lead to serious transmission damage if you overfill it based on that false reading.

The process is specific. After driving the car to warm up the fluid, park on a level surface, leave the engine running, and shift the transmission through each gear, pausing briefly before returning to 'Park'. Then, with the engine still idling, pull the transmission dipstick, wipe it clean, reinsert it fully, and pull it out again to get the true level. The fluid should be between the "Hot" marks on the dipstick.

The fluid expands when hot, and the pump needs to be working to get a real reading. If you check it cold with the engine off, you'll think it's low and might overfill it.
